African swine fever (ASF) is a highly infectious and fatal haemorrhagic disease of pigs that is caused by a DNA virus of the genus Asfivirus and Asfarviridae African suids family. Since September 2019, ASF has caused extensive mortalities to the pig population of Timor-Leste, where pigs are important for traditional ceremonies and for monetary income. To assist Timor-Leste in the management of this disease, we have developed a comprehensive multiplexed set of tiling primers adapted for the Oxford Nanopore (ONT) Rapid Barcoding library kit that allows rapid sequencing of the ASF virus (ASFV) genome. Different combinations of the primers allow for either whole-genome sequencing and/or targeted sequencing of the common gene markers currently used for the typing of ASFV. These primers were tried on two clinical ASFV samples in Timor-Leste in July 2024 and we will demonstrate here, the effectiveness of the developed tiling primers when applied to whole-genome and targeted sequencing of the virus on the ONT platform.
